R788 for RA Goes to Phase IIb Clinical Trial

Two Phase IIb clinical trials of the orally available Syk inhibitor, R788, are being initiated in people with rheumatoid arthritis (RA). The trials will evaluate the efficacy of R788 (fostamatinib disodium) compared with placebo. The trials are studying patients who have failed to respond to methotrexate or at least one biologic agent. Results are expected to be available late 2009.
INCB18424 Demonstrates Clinical Benefits in RA

Results from a Phase IIa trial of INCB18424, an orally available janus-associated kinase inhibitor, in patients with rheumatoid arthritis (RA) have been released. Results from the first of four treatment groups demonstrated that the 15-mg, twice-daily dose was well tolerated and provided positive response rates. Three additional groups are being evaluated. The results, coupled with the drug’s ability to block signaling from multiple inflammatory cytokines, suggest it has potential to be more effective than available RA therapies such as injectable biologics, the developing company said.
Second Phase 2/3 Lupus Study Underway

A Phase 2/3 trial of atacicept in people with systemic lupus erythematosus (SLE) has started. The one-year, randomized, double-blind, placebo-controlled, international study in 500 patients with SLE will evaluate the efficacy and safety of atacicept in preventing SLE flares.
A second Phase II/III study in lupus nephritis, a severe form of SLE that affects the kidneys, began last December.
